we describe the optical synapse, an approach to control synaptic connectivity that utilizes presynaptically originating bioluminescence to activate optogenetic actuators expressed at postsynaptic sites (fig. 1d). we have shown that bioluminescent optogenetics can operate within a cell, with a luciferase tethered to an opsin (a luminopsin) 6, 7, 8, 9 . photon release from the luciferin-luciferase interaction activates the associated opsin thereby achieving optogenetic modulation. depending on the biophysical properties of the opsin, excitatory or inhibitory, bioluminescence may depolarize or hyperpolarize the neuron.
in summary, interluminescence provides a unique technology for interrogating specific neural circuits with substantial temporal and spatial control. interluminescence can boost or down regulate synaptic efficacy at specific synapses, it can be used to bias synaptic output, e.g. from inhibitory to excitatory and vice versa, and, in principle, it can establish new functional synaptic connections for example from silent to active. with rapid advances in the available palette of luciferases and opsins, this strategy can expand to meet a wide array of experimental needs.
recording the activity of a single neuron with high spatial resolution over long periods of time is a major challenge in neuroscience. the few available methods are limited to only a few cells at a time, which usually requires extensive surgery. in contrast, light can be used to visualize activity in the brain non-invasively. light can enter and spread throughout the tissue, and thus can be used to trace the activity of many cells simultaneously. in this paper we describe light-sensitive optogenetics, a light-induced circuit control method that enables the control of single neurons. optogenetics refers to the genetic targeting of cells by the light-activated depolarization of ion channels. it is based on the natural light sensitivity of ion channels and uses light to control the activity of neurons. in this study we demonstrate the spatial and temporal control of large groups of neurons using light. we show that the light-induced depolarization of neurons can be used to control the activity of downstream neurons, control the development of synapses, and induce network oscillations. we demonstrate the potential of light-induced optogenetics for the control of single neurons in acute brain slices and in vivo. these experiments show that light-induced optogenetics can be used to control the activity of single neurons over time periods of hours in vivo.

sometimes it might seem like hydra is a faceless evil empire, but there are people, organizations, and even countries which support them in secret. the hydra organization itself is comprised of various individuals and organizations that share hydra ideologies, which can lead to a lot of overlap in leadership. while there are a number of regions or locations which have adopted hydra as their own, many of these have many members and factions. as a result, the overall structure of hydra has a lot of overlap and sometimes parts or factions are not always loyal to the overall hydra organization.
hydra is a powerful and sinister organization and you have to be dedicated to one of its ideals to join. these ideals can be as simple as the idea that you must serve the most powerful leader of hydra or as complex as you must follow a specific ideological path. those who don’t serve a single leader can be agents of hydra, but they can also be individuals who simply follow their own ideals. if you’re interested in joining hydra, it’s a good idea to study the organization and its goals and ideals.
